==== Mechanism of action ==== The mechanism of action of biguanides is not fully understood, and many mechanisms have been proposed for metformin. Biguanides do not affect the output of insulin, unlike other hypoglycemic agents such as sulfonylureas and meglitinides. Therefore, they are effective in Type 2 diabetics; and in Type 1 diabetes when used in conjunction with insulin therapy. == Sustainability == The effects of climate change are becoming more of a concern for organizations, and mitigation strategies are being sought by the research community. While many laboratories are used to perform research to find innovative solutions to this global challenge, sustainable working practices in the labs are also contributing factors towards a greener environment. Many labs, such as those at the Massacusetts Institute of Technology and the University of Edinburgh, are already trying to minimize their environmental impact by reducing energy consumption, recycling, and implementing waste sorting processes to ensure correct disposal. === Tendon-bone interface === In addition to its role in load-bearing joints, cartilage serves a crucial function as a gradient material between softer tissues and bone. Mechanical gradients are crucial for the body's function, and for complex artificial structures including joint implants. Interfaces with mismatched material properties lead to areas of high stress concentration which, over the millions of loading cycles experienced by human joins over a lifetime, would eventually lead to failure. == Pharmacology == The drug showed antidepressant-like and anxiolytic-like effects as well as locomotor-stimulating effects in animal models. It had reduced induction of seizures and locomotor hyperactivity compared to other δ-opioid receptor agonists. The doses required for stimulant-like activity were 3- to 10-fold greater than the doses that produced antidepressant- and anxiolytic-like effects. The drug appears to have a very low misuse potential based on animal studies. In addition to its δ-opioid receptor agonist activity, AZD-2327 has been reported to act as a cytochrome P450 CYP3A4 inhibitor. It has been found to inhibit the release of norepinephrine caused by anxiety and was able to do so as much as the benzodiazepine diazepam. However, AZD-2327 could be advantageous to benzodiazepines because these drugs often cause rapid tolerance and dependence. In contrast to benzodiazepines, AZD-2327 may have less or no potential for tolerance in terms of its anxiolytic-like effects.
