Tesamorelin binds to growth hormone-releasing hormone receptors on somatotroph cells in the anterior pituitary. Receptor activation increases intracellular cyclic AMP and promotes synthesis and secretion of growth hormone. Because the peptide mimics endogenous GHRH, it amplifies the normal pulsatile release of growth hormone rather than providing exogenous growth hormone directly. This upstream action distinguishes tesamorelin from recombinant growth hormone preparations and from growth hormone secretagogues that act at different receptors.
Stimulated growth hormone release leads to hepatic production of insulin-like growth factor 1, a key mediator of many growth hormone effects. In clinical studies, tesamorelin increased IGF-1 levels in a dose-dependent manner, although the response varies among individuals. The drug's effect on visceral fat is thought to involve growth hormone-mediated lipolysis and altered adipocyte metabolism. Muscle mass and lean body mass have also been assessed as secondary outcomes, but changes are generally smaller and less consistent than fat reductions.
Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.

Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from truncated or oxidized forms. Mass spectrometry confirms the expected molecular weight, and peptide mapping after enzymatic digestion verifies the amino acid sequence. Water content is measured because residual moisture affects stability, and tests for aggregates or particulates are standard for injectable peptides. Circular dichroism can indicate whether the molecule has adopted an unexpected secondary structure in solution.
Research supply is often accompanied by a certificate of analysis listing chromatographic purity, mass confirmation, and storage conditions. Laboratories compare that document with an independent test when material is intended for bench work, since certificates describe a batch rather than an individual vial. Published studies usually state the source and purity of the peptide because small differences in purity can shift measured activity. Full analytical validation is rarely reported, which leaves batch-to-batch comparability an open question.
The peptide is supplied as a lyophilized powder in single-use vials and is normally kept refrigerated between two and eight degrees Celsius, protected from light. Once dissolved, the solution is handled carefully because peptide bonds and the acyl modification can degrade under warm or alkaline conditions. Vials are inspected for cracks, and the powder is checked for color and uniformity before handling. Temperature excursions during shipping are a frequent reason for quality questions.
==== Alternative theories ==== The 5,000-year-old mummified body of Ötzi the Iceman was found with 15 groups of tattoos, many of which were located at points on the body where acupuncture needles are used for abdominal or lower back problems. Evidence from the body suggests Ötzi had these conditions. This has been cited as evidence that practices similar to acupuncture may have been practised elsewhere in Eurasia during the early Bronze Age; however, The Oxford Handbook of the History of Medicine calls this theory "speculative". It is considered unlikely that acupuncture was practised before 2000 BC. Acupuncture may have been practised during the Neolithic era, near the end of the Stone Age, using sharpened stones called Bian shi. Many Chinese texts from later eras refer to sharp stones called "plen", which means "stone probe", that may have been used for acupuncture purposes. The ancient Chinese medical text, Huangdi Neijing, indicates that sharp stones were believed at-the-time to cure illnesses at or near the body's surface, perhaps because of the short depth a stone could penetrate. However, it is more likely that stones were used for other medical purposes, such as puncturing a growth to drain its pus. The Mawangdui texts, which are believed to be from the 2nd century BC, mention the use of pointed stones to open abscesses, and moxibustion, but not for acupuncture. It is also speculated that these stones may have been used for bloodletting, due to the ancient Chinese belief that illnesses were caused by demons within the body that could be killed or released.
Denaturing High Performance Liquid Chromatography (DHPLC) is a method of chromatography for the detection of base substitutions, small deletions or insertions in the DNA. Due to its speed and high resolution, this method is particularly useful for finding polymorphisms in DNA. In practice, the analysis begins with a standard polymerase chain reaction (PCR) in order to amplify the fragment of interest. If the amplified region that exhibits the polymorphism(s) is heterozygous, two kinds of fragments corresponding to the allele and the wild polymorphic allele will be present in the PCR product. This first step is followed by a step of denaturation–renaturation to create hetero- and homoduplexes from the two allele populations in the PCR. To find a homozygous polymorphism, proceed in the same way by premixing a DNA wild population to a population of polymorphic DNA to obtain heteroduplexes after the denaturation–renaturation step. Heteroduplexes are actually double strands of DNA containing a strand from the wild-type allele and a sprig from the polymorphic allele. The formation of such DNA fragments then causes the appearance of a "mismatch" or bad pairing where the polymorphism is located. These "mismatches" in the heteroduplex are the basis for the polymorphism detection by DHPLC. Heteroduplexes are thermally less stable than their corresponding homoduplexes, and the single DNA strands will therefore be disconnected by chromatography when subjected to a sufficiently high temperature.
It uses a wide range of organic material for food; in animals, its versatility enables it to infect damaged tissues or those with reduced immunity. The symptoms of such infections are generalized inflammation and sepsis. If such colonizations occur in critical body organs, such as the lungs, urinary tract, and kidneys, the results can be fatal. Because it thrives on moist surfaces, this bacterium is also found on and in soap and medical equipment, including catheters, causing cross-infections in hospitals and clinics. It is also able to decompose hydrocarbons and has been used to break down tarballs and oil from oil spills. P. aeruginosa is not extremely virulent in comparison with other major species of pathogenic bacteria such as Gram-positive Staphylococcus aureus and Streptococcus pyogenes , although P. aeruginosa is capable of extensive colonization and can aggregate into enduring biofilms. Its genome includes numerous genes for transcriptional regulation and antibiotic resistance, such as efflux systems and beta-lactamases, which contribute to its adaptability and pathogenicity in human hosts. P. aeruginosa produces a characteristic sweet, grape-like odor due to its synthesis of 2-aminoacetophenone.

Cachexia ( ) is a syndrome that occurs in people with certain illnesses, causing muscle loss that cannot be fully reversed with improved nutrition. It most commonly occurs in cases of cancer, congestive heart failure, chronic obstructive pulmonary disease, chronic kidney disease, and AIDS. These conditions change how the body handles inflammation, metabolism, and brain signaling. This can lead to muscle loss and other harmful changes to body composition over time. Unlike weight loss from inadequate caloric intake, cachexia mainly causes muscle loss and can happen with or without fat loss. Diagnosis of cachexia is difficult because there are no clear guidelines, and its occurrence varies from one affected person to the next. Like malnutrition, cachexia can lead to worse health outcomes and lower quality of life. The prognosis of patients with cachexia varies depending on the type and severity of the underlying illness, but is typically poor, especially with patients in late stages of disease. Cachexia can improve significantly with effective treatment of the underlying illness, but symptomatic treatment approaches such as nutritional therapy and exercise typically do not result in reversal of the syndrome, and have very limited benefit in advanced cases of cachexia.

==== The Buddha's discovery of conditionality ==== Regarding the arising of suffering, SN 12.10 discusses how before the Buddha's awakening, he searched for the escape from suffering as follows: "when what exists is there old age and death? What is a condition for old age and death?", discovering the chain of conditions as expressed in the twelve nidanas and other lists. MN 26 also reports that after the Buddha's awakening, he considered that dependent origination was one of the two principles which were "profound (gambhira), difficult to see, difficult to understand, peaceful, sublime, beyond the scope of mere reasoning (atakkāvacara), subtle." The other principle which is profound and difficult to see is said to be Nirvana, "the stopping, or transcending, of conditioned co-arising" (Harvey). In the Mahānidānasutta (DN 15) the Buddha states that dependent origination is "deep and appears deep", and that it is "because of not understanding and not penetrating this teaching" that people become "tangled like a ball of string" in views (diṭṭhis), samsara, rebirth and suffering. SN 12.70 and its counterpart SA 347 state that "knowledge of Dhamma-stability" (dhamma-tthiti-ñānam) comes first, then comes knowledge of nirvana (nibbane-ñānam). However, while the process which leads to nirvāna is conditioned, nirvāna itself is called "unborn, unbecome, unmade, unconstructed" (Ud. 80–1). The Milinda Panha compares to how a mountain is not dependent on the path that leads to it (Miln. 269)". According to Harvey, since it is "not co-arisen (asamuppana) (It.
Integrative, multi-locus studies have prompted major revisions of several lichen families and genera. Two examples illustrate the impact of these studies: the re-circumscription of the family Graphidaceae and of the family Ramalinaceae. Graphidaceae—the script lichen family—contains more than 2,000 mostly tropical species. Traditional taxonomy centred on fruiting body shape and a few microscopic characters. A five-locus phylogeny (Rivas-Plata and Lumbsch 2011) showed that many characters had evolved convergently. Slit-like versus rounded discs arose repeatedly, so genera built on those traits mixed unrelated species. Molecular results led to a wholesale recircumscription: several traditional genera were divided and new ones erected to reflect monophyletic clades. For instance, the broad Graphis was split into several smaller, genetically and chemically homogeneous genera. This process was facilitated by projects such as Ticolichen (a tropical lichen inventory project) which combined fieldwork, morphology, chemistry, and DNA sequencing to tackle these revisions. The outcome is a more natural scheme: genera now align with monophyletic clades, even when outwardly dissimilar species must be grouped together. The revision greatly increased recognized genera and species, exposing hidden diversity in tropical crustose lichens. Another major revision, in this case of a mostly temperate/tropical group, was carried out by Kistenich and colleagues (2018) on the family Ramalinaceae. The family had about 40 genera of uncertain affinity, several delimited by only one or two traits.

According to Reuters, the conflict involving Iran has resulted in increased oil prices, elevated inflation rates, and a subsequent rise in the cost of living within the United States. Furthermore, a majority of voters in 2025 report experiencing a decline in their economic well-being under the administration of President Trump.
The British historian Richard Overy wrote the huge demands of the Four Year Plan "...could not be fully met by a policy of import substitution and industrial rationalisation", thus leading Hitler to decide in November 1937 that to stay ahead in the arms race with the other powers that Germany had to seize Czechoslovakia in the near-future. At the Hossbach conference on 5 November 1937, Hitler announced that seizing Czechoslovakia would increase the supply of food under German control, which in turn would lessen the need to import food, thereby freeing up more foreign exchange to import raw materials necessary for the Four Year Plan's targets. The Hossbach conference was largely taken up with an extended discussion about the necessity of bringing areas adjunct to Germany under German economic control, by force if necessary, as Hitler argued that this was the best way to win the arms race. Hitler stated: "areas producing raw materials can be more usefully sought in Europe, in immediate proximity to the Reich". Overy wrote about Hitler's attitude to the Reich's economic problems that: "He simply saw war instrumentally, as the Japanese had done in Manchuria, as a way to expand the German resource base and to secure it against other powers". At the time, Czechoslovakia had Europe's 7th largest economy, and easily the most modern, developed, and industrialized economy in Eastern Europe.
=== Physical characteristics === Diallyl disulfide has a strong garlic smell. It is a clear, yellowish liquid which boils at 138–139 °C (for the typical 80% purity) and has its flash point at 50 °C, a density of about 1.0 g/mL and a vapor pressure of 1 mmHg at 20 °C. It is non-polar; therefore, diallyl disulfide is insoluble in water and is soluble in fats, oils, lipids, and non-polar solvents such as hexane or toluene.
