Kruger re-equipped the Transvaal army, importing 37,000 of the latest 7x57 mm Mauser Model 1895 rifles supplied by Germany, and 40 to 50 million rounds of ammunition. Some commandos used the Martini-Henry Mark III, because thousands of these had been purchased. Unfortunately, the large puff of white smoke after firing gave away the shooter's position. Roughly 7,000 Guedes 1885 rifles had also been purchased a few years earlier, and these were also used during the hostilities. As the war went on, some commandos relied on captured British rifles, such as the Lee-Metford and Enfield. When the ammunition for the Mausers ran out, the Boers relied primarily on the captured Lee-Metfords. Few Boers used bayonets. The Boers also purchased the best European German Krupp artillery. By October 1899, the Transvaal State Artillery had 73 heavy guns, including four 155 mm Creusot fortress guns and 25 of the 37 mm Maxim Nordenfeldt guns. The Boers' Maxim, larger than the British Maxims, was a large calibre, belt-fed, water-cooled "auto cannon" that fired explosive rounds at 450 rounds per minute. It became known as the "Pom Pom". The Transvaal army was transformed: approximately 25,000 men equipped with modern rifles and artillery could mobilise within two weeks. However, Kruger's victory in the Jameson Raid did nothing to resolve the fundamental problem of finding a formula to conciliate the uitlanders, without surrendering the independence of the Transvaal.
== Disorder and disease == Intrinsically unstructured proteins have been implicated in a number of diseases. Aggregation of misfolded proteins is the cause of many synucleinopathies and toxicity as those proteins start binding to each other randomly and can lead to cancer or cardiovascular diseases. Thereby, misfolding can happen spontaneously because millions of copies of proteins are made during the lifetime of an organism. The aggregation of the intrinsically unstructured protein α-synuclein is thought to be responsible. The structural flexibility of this protein together with its susceptibility to modification in the cell leads to misfolding and aggregation. Genetics, oxidative and nitrative stress as well as mitochondrial impairment impact the structural flexibility of the unstructured α-synuclein protein and associated disease mechanisms. Many key tumour suppressors have large intrinsically unstructured regions, for example p53 and BRCA1. These regions of the proteins are responsible for mediating many of their interactions. Taking the cell's native defense mechanisms as a model drugs can be developed, trying to block the place of noxious substrates and inhibiting them, and thus counteracting the disease.
==== Association with insulin secretory granules ==== Much, but not all, of the glucokinase found in the cytoplasm of beta cells is associated with insulin secretory granules and with mitochondria. The proportion thus "bound" falls rapidly in response to rising glucose and insulin secretion. It has been suggested that binding serves a purpose similar to the hepatic glucokinase regulatory protein—protecting glucokinase from degradation so that it is rapidly available as the glucose rises. The effect is to amplify the glucokinase response to glucose more rapidly than transcription could do so.
=== 2011: Rebuilding and named the Army Reserve === In 2010, the government set up a commission, chaired by General Sir Nicholas Houghton, to review the state of the reserves and to design their future. The commission reported in July 2011. It found that, despite their operational commitments, the reserves had been neglected: some estimates put its trained and active strength as low as 14,000. There had been 'a failure to resource recruiting and good training, especially collective training; to offer career progression; to update operational roles; to permit deployment in formed sub-units and therefore offer command opportunities' Its recommendations included that a new target of 30,000 trained strength by 2020 be set. The government published the report on 18 July with an immediate promise of £1.5 billion funding over ten years. In October 2012 MoD announced a new name for the Territorial Army, its current name, the Army Reserve. Most of the recommendations of the commission were adopted in the White Paper July 2013 including the critical importance of providing operational and training opportunities for formed units and sub-units. Some aspects required legislation and were introduced in the Defence Reform Act (2014). These included the name change, reforms in mobilisation arrangements and in employer compensation.
