== Sample preparation == Sample preparation is one of the most critical steps in NanoSIMS analysis, particularly when analysing biological samples. Specific protocols should be developed for individual experiments in order to best preserve not only the structure of the sample but also the true spatial distribution and abundance of molecules within the sample. As the NanoSIMS operates under ultra high vacuum, the sample must be vacuum compatible (i.e., volatile free), flat, which reduces varying ionization trajectories, and conductive, which can be accomplished by sputter coating with Au, Pt, or C. Biological samples, such as cells or tissue, can be prepared with chemical fixation or cryo-fixation and embedded in a resin before sectioning into thin slices (100 nm - 1μm), and placed on silicon wafers or slides for analysis. Sample preparation for metallographic samples is generally much simpler but a very good metallographic polish is required to achieve a flat, scratch free surface.

== Medical uses == Setmelanotide is indicated for chronic weight management (weight loss and weight maintenance for at least one year) in people six years and older with obesity due to three rare genetic conditions: pro-opiomelanocortin (POMC) deficiency, proprotein subtilisin/kexin type 1 (PCSK1) deficiency, and leptin receptor (LEPR) deficiency confirmed by genetic testing demonstrating variants in POMC, PCSK1, or LEPR genes considered pathogenic (causing disease), likely pathogenic, or of uncertain significance. Setmelanotide is the first FDA-approved treatment for these genetic conditions. Setmelanotide is not approved for obesity due to suspected POMC, PCSK1, or LEPR deficiency with variants classified as benign (not causing disease) or likely benign or other types of obesity, including obesity associated with other genetic syndromes and general (polygenic) obesity.
== Preparation == An aqueous solution of sodium silicate is acidified to produce a gelatinous precipitate that is washed, then dehydrated to produce colorless silica gel. When a visible indication of the moisture content of the silica gel is required, ammonium tetrachlorocobaltate(II) (NH4)2[CoCl4] or cobalt(II) chloride CoCl2 is added. This will cause the gel to be blue when dry and pink when hydrated. Due to a link between the cobalt chloride and cancer, it has been forbidden in Europe for use in silica gel. An alternative indicator is methyl violet, which is orange when dry and green when hydrated.

=== Antibiotics === The length of antibiotic courses depends on the severity of the infection and whether bone infection is involved, but can range from 1 week to 6 weeks or more. Current recommendations are that antibiotics are only used when there is evidence of infection and continued until there is evidence that the infection has cleared, instead of evidence of ulcer healing. An example of private sector whistleblowing is when an employee reports to someone in a higher position such as a manager or to external factors, such as their lawyer or the police. Whistleblowing in the private sector is typically not high-profile or openly discussed in major news outlets, though occasionally, third parties expose human rights violations and exploitation of workers. Many governments attempt to protect such whistleblowers. In the United States, for example, there are organizations such as the United States Department of Labor (DOL) and laws such as the Sarbanes-Oxley Act and the United States Federal Sentencing Guidelines for Organizations (FSGO) that protect whistleblowers in the private sector.
