=== United States === Tincture of Opium is available by prescription in the United States. It is regulated as a Schedule II drug (No. 9639) under the Controlled Substances Act. In the United States, opium tincture is marketed and distributed by several pharmaceutical firms, each producing a single formulation of the drug, which is deodorized. Each mL contains 10 mg of anhydrous morphine (the equivalent of 100 mg of powdered opium), other opium alkaloids (except noscapine), and ethanol, 19%. It is available packaged in bottles of four US fluid ounces (118 mL) and 16 US fluid ounces (1 US pt; 473 mL). Tincture of Opium is known as one of many "unapproved drugs" regulated by the U.S. Food and Drug Administration (FDA); the marketing and distribution of opium tincture prevails only because opium tincture was sold prior to the Federal Food, Drug & Cosmetic Act of 1938. Its "grandfathered" status protects opium tincture from being required to undergo strict FDA drug reviews and subsequent approval processes. However, the FDA closely monitors the labeling of opium tincture. Bottles of opium tincture are required by the FDA to bear a bright red "POISON" label given the potency of the drug and the potential for overdose (see discussion about confusion with Paregoric below). Additionally, in a warning letter to a manufacturer of opium tincture in late 2009, the FDA noted that "we found that your firm is manufacturing and distributing the prescription drug Opium Tincture USP (Deodorized – 10 mg/mL).
Susineridine (INNTooltip International Nonproprietary Name; developmental code name YZJ-4729) is an atypical μ-opioid receptor (MOR) agonist and opioid analgesic which is under development for the treatment of postoperative pain. It is given intravenously. The drug acts as a selective biased partial agonist of the MOR, with functional selectivity for activation of G protein signaling over β-arrestin2 recruitment. Its mechanism of action and pharmacological activity are said to be similar to those of oliceridine. However, in-vitro findings suggest that susineridine may have a greater degree of G protein bias than oliceridine. The drug produces analgesic effects in multiple animal models of pain and produces less respiratory depression than morphine or oliceridine. The pharmacokinetics of susineridine in humans have been studied. Susineridine was patented and first described in the scientific literature in 2023. It is under development by Shanghai Haiyan Pharmaceutical Technology in China. As of May 2025, the drug is in phase 3 clinical trials for postoperative pain.
=== Grhl3 === Much the same as the previous two orthologues, Grhl3 is involved in the regulation of epidermal tissue, such as the formation of the jaw, neural tube and other craniofacial features, and does so across both land and aquatic organisms. Grhl3 is a downstream target of Irf6, and plays a key role in processes involving fusion during development much like Grhl2, especially so in the oral palate and spinal cord. A mutation of Grhl3, that causes an increase or a decrease in expression, can lead to Van der Woude syndrome, which is characterized by phenotypes that include cleft lip and/or palate and spina bifida. Primarily, Grhl3 appears to play a vital role for regulating the development of the craniofacial skeleton. A genome-wide association study found that Grhl3 is an etiological variant for a nonsyndromic form of cleft palate, ~50% of all cleft palate cases, highlighting the level of impact that dysregulation of Grhl3 has on development. Apart from the defects that are physically noticeable, Grhl3 is also expressed in the brain of mice embryos and has been shown to regulate the impulsiveness and anxiety levels of mice. Furthermore, it appears that grhl3 regulates the enveloping layer of zebrafish and axial extension as well as cell size and identity during embryonic development. If expression is disrupted during the early stages of disruption it will lead to severe defects that can lead to the death of the embryo before epiboly is complete.

Eladocagene exuparvovec is an adeno-associated virus vector-based gene therapy. The most common side effects include initial insomnia, irritability and dyskinesia. The most common side effects of eladocagene exuparvovec (Kebilidi) include dyskinesia (involuntary muscle movements), fever, low blood pressure, anemia (low red blood cell count), increased saliva production, insomnia, low levels of potassium, phosphate, and/or magnesium, and procedural complications such as respiratory and cardiac arrest. In February 2024, the American Psychological Association approved a policy statement supporting unobstructed access to health care and evidence-based clinical care for transgender, gender-diverse, and nonbinary children, adolescents, and adults, as well as opposing state bans and policies intended to limit access to such care. In May 2025, a two-year systematic review commissioned by the state of Utah as part of a ban on trans youth healthcare concluded that "The consensus of the evidence supports that the treatments are effective in terms of mental health, psychosocial outcomes, and the induction of body changes consistent with the affirmed gender in pediatric [gender dysphoria] patients. The evidence also supports that the treatments are safe in terms of changes to bone density, cardiovascular risk factors, metabolic changes, and cancer".
